The partition coefficient (Log P) value was defined as the ratio of unionized drugs partitioned between the aqueous and organic phases. The oil-water partition coefficient reflects the ability of a drug to cross a lipid membrane. The lipophilic/hydrophilic balance is one of the most important influencing factors for optimal drug absorption and delivery. Due to the lipid nature of biological membranes, the amount of drug absorbed depends largely on their lipophilicity.

A Log P value of 0 indicates that the drug is evenly distributed in both the water and the partition solvent. Log P values less than 1 indicate higher water solubility, while values greater than 1 indicate higher fat solubility. For optimal solubility and absorption, a proper hydrophilic-lipophilic balance is necessary.

Shake flask method | The shake flask method is to shake a flask containing two immiscible solvents and a solute. After reaching equilibrium, analyze the concentration of the solute in one or both phases, thereby calculating logP. The shake flask method usually utilizes an octanol-water system to determine LogP. Octanol was chosen as the partitioning solvent for several reasons:
Octanol is thought to mimic the lipidic properties of biological membranes because it contains a polar head and a nonpolar tail.
Octanol is an organic compound that is immiscible in water.
Most drugs have solubility parameters similar to octanol. |
RP-HPLC | Reverse-phase high-performance liquid chromatography has the advantages of simple operation, rapidity, low cost, and good repeatability, and is a good method for indirect determination of drug logP. |
